Involvement of Lipin-1 in TLR4 Signaling
نویسنده
چکیده
Our lab is focused on the study of a family of proteins known as lipins that exhibit phosphatidic acid phosphatase activity, and the role they play in macrophages. Up to date 5 proteins have been described that are encoded by three different genes. The lpin1 gene may undergo alternative splicing. generating lipin1α, β and γ isoforms. Lipin1γ is present only in humans. These proteins have a very similar protein structure. They have two conserved domains: the N-LIP in their n-terminal part, and the C-LIP in their cterminal end, where the catalytic and transcriptional coactivator motifs are located. They also have a nuclear localization signal in the N-terminal part.

The protective effect of nano-curcumin in experimental model of acute pancreatitis: The involvement of TLR4/NF-kB pathway
Objective(s): The objective of the present study is to explore whether Nanocurcumin improves pancreatic inflammation through the inhibition of the TLR4/NFkB signaling pathway in cerulein-induced acute pancreatitis. Methods: Acute pancreatitis was induced by five intraperitoneal (i.p.) injection of cerulein (50 μg/kg) with 1h intervals. Vehicle and nanocurcumin (100mg/kg/day) were given to the a...

Using macrophages from lipin-1-deficient animals and human macrophages deficient in the enzyme, we have found that this phosphatase acts as a proinflammatory mediator during TLR signaling and during the development of in vivo inflammatory processes. After TLR4 stimulation lipin-1 deficient macrophages showed a decreased production of diacylglycerol, activation of mitogenactivated protein kinase...
متن کاملLipin-1 integrates lipid synthesis with proinflammatory responses during TLR activation in macrophages.
Lipin-1 is a Mg(2+)-dependent phosphatidic acid phosphatase involved in the de novo synthesis of phospholipids and triglycerides. Using macrophages from lipin-1-deficient animals and human macrophages deficient in the enzyme, we show in this work that this phosphatase acts as a proinflammatory mediator during TLR signaling and during the development of in vivo inflammatory processes. After TLR4...
متن کاملTLR2 and TLR4 Signaling Pathways and Gastric Cancer: Insights from Transcriptomics and Sample Validation
Background: Pattern recognition receptors, especially toll-like receptors (TLRs), as the first line of defense for pathogen detection, were found to be associated with H. pylori infection and gastric cancer (GC). However, the expression levels of TLRs, i.e. TLR2 and TLR4, as the main receptors sensed by H. pylori, still remain largely ambiguous. We aimed to investigate the patterns of key tra...
متن کاملFiltered Kombucha Tea Rings the Bell for TLR2, TLR4, MYD88, and Dectin-1 in Mice Model of Colitis
Background and objectives: TLR2, TLR4, and Dectin-1 (Clec7) are pattern recognition receptors (PRRs) expressed by intestinal epithelia cells and MYD88 is a signaling molecule of TLR2 and TLR4. They warn immune system about the presence of invading pathogens promoting initiation of inflammatory response. Because of colonic cancer risk, therapy of intestinal inflammation is of h...
